First Chromosome-Scale Human Genome Assembly from Karnataka
This landmark study presents the first chromosome-level de novo genome assembly of an individual from Karnataka, India (KIn1), generated using PacBio HiFi long-read sequencing integrated with Hi-C chromatin scaffolding. The highly contiguous assembly enabled high-resolution reconstruction of chromosomal architecture and comprehensive characterization of complex genomic regions.
Comparative analysis with globally diverse human genomes identified population-specific structural variations, including unique inversions and chromosomal translocations, highlighting the distinct genomic landscape of South Asian populations. The study also demonstrated strong genetic proximity between KIn1 and South Asian ancestry groups such as ITU and STU, emphasizing the importance of ethnicity-specific reference genomes for population genomics, structural variant discovery, and precision medicine applications in India.
Significance in Human Genomics & Precision Medicine
This study establishes a high-resolution genomic framework for understanding South Asian genetic diversity through chromosome-scale human genome assembly. By uncovering ethnicity-specific structural variants and complex chromosomal rearrangements, the research advances population genomics, ancestry mapping, and precision medicine applications in Indian populations.
The findings further highlight the power of long-read genome assembly technologies in enabling accurate characterization of rare variants and previously unresolved genomic regions beyond the limitations of conventional reference genomes.
